Pre-race favourites Kevin Evans and David George from Team MTN, blasted away from the rest of the field to win the prologue stage, 17 kilometres at the Pezula Golf Estate, by 57 seconds.
Hardly any climbing today, but tomorrow's first stage from Knysna to George will see 3091 metres of climbing.
The ladies Trek team of Americans Susan Haywood and Jennifer Smith have also grabbed a lead.
The 8 stage race finishes at Lourensford, in Somerset West, next week Saturday.
